PEG衍生物與蛋白藥物修飾和PEG材料介紹
藥物的聚乙二醇修飾即聚乙二醇化,是將活化的聚乙二醇通過(guò)化學(xué)方法偶聯(lián)到蛋白、多肽、小分子有機(jī)藥物和脂質(zhì)體上是一種蛋白質(zhì)工程分子修飾方法。藥物的聚乙二醇修飾可分為兩個(gè)階段。
*階段的修飾技術(shù)局限于應(yīng)用相對(duì)分子質(zhì)量低的單甲氧基聚乙二醇(<20000)。常用的修飾劑有單甲氧基聚乙二醇琥珀酸琥珀酰亞胺酯(mPEG-SS)、單甲氧基聚乙二醇碳酸琥珀酰亞胺酯(mPEG-SC)等,通過(guò)酯鍵或三嗪環(huán)將聚乙二醇與藥物分子偶聯(lián),這種非特異性的不穩(wěn)定連接方式使得一個(gè)藥物分子經(jīng)常連接數(shù)個(gè)聚乙二醇分子。但*代聚乙二醇修飾藥物通常表現(xiàn)出不穩(wěn)定性、較大的毒性和免疫原性,生物活性、藥代動(dòng)力學(xué)的性質(zhì)與原型藥物沒(méi)有本質(zhì)的改變。
以應(yīng)用相對(duì)分子質(zhì)量高(>20000)聚乙二醇修飾劑為特征的第二階段的修飾技術(shù)具有連接穩(wěn)定、定點(diǎn)修飾、控釋等特點(diǎn),修飾后的藥物具有更高的生物活性、更好的物理及熱穩(wěn)定性、更高的產(chǎn)品均一性和純度。
但總得說(shuō)來(lái),蛋白藥物PEG修飾技術(shù)中zui大的問(wèn)題就是無(wú)法實(shí)現(xiàn)定點(diǎn)修飾,修飾產(chǎn)物不均一,給分離純化帶來(lái)很大難度,也很大程度上阻礙了臨床應(yīng)用。根據(jù)蛋白質(zhì)的氨基酸性質(zhì)和PEG衍生物的特點(diǎn),科研人員開(kāi)發(fā)了多種定點(diǎn)修飾的策略和方法。
有一些蛋白的N端對(duì)其生物活性起著重要作用,如果在N端實(shí)施PEG修飾則會(huì)使蛋白的生物活性喪失,因此將PEG修飾的位點(diǎn)轉(zhuǎn)移至C端則是一種有效的修飾策略。一般選用mPEG-酰肼或mPEG-胺對(duì)蛋白質(zhì)進(jìn)行修飾,羧基的修飾位點(diǎn)包括天冬氨酸、谷氨酸及末端羧基。mPEG-酰肼是近年來(lái)開(kāi)發(fā)的又一種可與羧基特異性結(jié)合的修飾劑。
與賴氨酸相比,半胱氨酸在蛋白質(zhì)中出現(xiàn)幾率和數(shù)量很少,利用具有巰基反應(yīng)活性PEG衍生物就可以實(shí)現(xiàn)蛋白定點(diǎn)修飾,但缺點(diǎn)是半胱氨酸的巰基通常處于蛋白的活性中心,修飾后會(huì)使蛋白活性損失較大。對(duì)于沒(méi)有半胱氨酸的蛋白則可以通過(guò)基因工程手段引入巰基反應(yīng)位點(diǎn)。這一方法不但能夠?qū)崿F(xiàn)高度選擇性修飾,而且能夠減少蛋白生物活性的喪失,并降低免疫原性。 目前可用于巰基修飾的mPEGs包括mPEG-馬來(lái)酰亞胺、mPEG-乙烯基砜及mPEG-碘乙酰胺等。其中mPEG-馬來(lái)酰亞胺制備簡(jiǎn)單,應(yīng)用較多。PEG-鄰吡啶-與蛋白質(zhì)偶聯(lián)形成的S—S鍵,在生理?xiàng)l件下可緩慢水解,釋放出天然蛋白質(zhì),有利于活性的恢復(fù),但修飾率低。PEG-乙烯基砜在pH7-9范圍內(nèi)可選擇性的與巰基反應(yīng),當(dāng)pH較高時(shí)則與氨基交聯(lián)。PEG-碘乙酰胺的優(yōu)勢(shì)在于經(jīng)強(qiáng)酸水解后,可釋放出半胱氨酸衍生物,易于鑒定。
對(duì)蛋白中含有的二硫鍵進(jìn)行PEG定點(diǎn)修飾分為2個(gè)步驟:首先利用溫和的還原劑使二硫鍵還原釋放兩個(gè)硫氫基;第二步通過(guò)PEG修飾劑的雙烷基化反應(yīng)重建二硫鍵,從而完成PEG的定點(diǎn)修飾。在修飾蛋白過(guò)程中不會(huì)出現(xiàn)蛋白的不可逆變性,而且二硫鍵還原后蛋白仍維持其三級(jí)結(jié)構(gòu),PEG選擇性的修飾兩個(gè)硫基,PEG的立體屏障作用確保1個(gè)二硫鍵只結(jié)合1分子PEG。但是二硫鍵PEG修飾可能會(huì)由于PEG的屏蔽作用,降低蛋白的生物活性。Veronese等利用變性劑鹽酸胍(3mol/L)使蛋白部分變性,結(jié)構(gòu)打開(kāi),但不使用還原劑以保護(hù)二硫鍵,在非折疊狀態(tài)下完成PEG修飾后,通過(guò)透析或凝膠過(guò)濾色譜的方法去除變性劑,使蛋白重新正確折疊恢復(fù)生物活性通常只有具有極性的氨基酸殘基的側(cè)鏈基團(tuán)才能夠進(jìn)行PEG化學(xué)修飾。Deiters等開(kāi)發(fā)了一種定點(diǎn)修飾非極性氨基酸苯丙氨酸的方法,首先用疊氮化物通過(guò)親核取代反應(yīng)修飾蛋白質(zhì)中的苯丙氨酸生成對(duì)位疊氮化苯丙氨酸,再用炔烴衍生的PEG-酰胺修飾劑修飾苯丙氨酸,發(fā)生環(huán)化加成反應(yīng)形成穩(wěn)定的連接體,從而完成對(duì)蛋白的定點(diǎn)修飾。另外,通過(guò)基因重組在蛋白質(zhì)中引入含疊氮基團(tuán)的氨基酸殘基,再與特定PEG修飾劑偶聯(lián)形成穩(wěn)定的定點(diǎn)單修飾產(chǎn)物。Cazalis通過(guò)基因工程方法在血栓調(diào)節(jié)蛋白(Thrombomodulin)的C端引入疊氮蛋氨酸,與mPEG2三芳基膦(mPEG2triarylphosphine)修飾劑生成穩(wěn)定的單修飾物,盡管該方法比較復(fù)雜,但至少給我們提供了一個(gè)新的PEG修飾蛋白的思路
此類PEG定點(diǎn)修飾的原理是:首先用葡萄糖氧化酶或高磺酸鈉氧化糖類殘基產(chǎn)生具有反應(yīng)活性的醛基,這些醛基可與mPEG2肼衍生物反應(yīng)生成腙或與mPEG2胺衍生物反應(yīng)生成可逆的Schiff堿。用硼氫甲腈鈉可將腙還原為更穩(wěn)定的烷基肼化物,而Schiff堿可以還原為仲胺。相對(duì)于mPEG-胺衍生物修飾,PEG-肼衍生物修飾不會(huì)形成交聯(lián)聚合體,更適合于糖基的修飾,zui有代表性的mPEG2肼衍生物是mPEG2酰肼(hydrazide2mPEG)。酰肼基團(tuán)極低的pKa值使得酰肼基團(tuán)在酸性環(huán)境下(pH4.5~5.0)中仍可以和羧基結(jié)合,而蛋白質(zhì)中的氨基在該條件下由于發(fā)生質(zhì)子化而不能與羧基反應(yīng),從而可實(shí)現(xiàn)選擇性定位修飾。
由于蛋白種類多樣和結(jié)構(gòu)復(fù)雜,往往會(huì)使得PEG修飾反應(yīng)特異性不強(qiáng),反應(yīng)條件要通過(guò)大量試驗(yàn)優(yōu)化。如果能夠利用計(jì)算機(jī)技術(shù)對(duì)蛋白質(zhì)晶體結(jié)構(gòu)表面性質(zhì)進(jìn)行理論計(jì)算和分析,確定可能的反應(yīng)位點(diǎn)和基團(tuán),并根據(jù)被修飾基團(tuán)與修飾劑的結(jié)構(gòu)特點(diǎn)設(shè)計(jì)合適的連接物,這樣不但可以減少PEG修飾的盲目性,而且能夠提高研究效率。胡遠(yuǎn)東等利用InsightⅡ分子模擬軟件包(BiosymInc.,SanDiego,CA)對(duì)牛血紅蛋白晶體(bHb)進(jìn)行了結(jié)構(gòu)分析和分子模擬,計(jì)算了蛋白表面24個(gè)可進(jìn)行PEG修飾的Lys殘基及其主要原子的溶劑可及表面積,確定了其中可以進(jìn)行修飾的13個(gè)Lys殘基(溶劑可及表面積>0.4nm2),并進(jìn)一步設(shè)計(jì)了具有雙功能基團(tuán)的小分子化合物環(huán)氧乙烷基活化PEG作為連接體。結(jié)果表明,PEG修飾的bHb的攜氧能力和免疫原性等生物學(xué)指標(biāo)均達(dá)到預(yù)期結(jié)果。
