Cal 27 was established in 1982 by J. Gioanni (Centre Antoine Lacassagne, Nice Cedex, France) from tissue taken prior to treatment from a 56 year old Caucasian male with a lesion of the middle of the tongue. CAL 27 cells are epithelial, polygonal with a highly granular cytoplasm. Immunocytochemical studies show strong positive staining with anti keratin antibodies. The cells do not grow well in semi-solid medium. Marked inhibition of thymidine incorporation was observed in the presence of VP16 (etoposide), CCNU (1-[2-chloroethyl]-3-cyclohexyl-1-nitrosourea), VM26 (teniposide), ADM (adriamycin), CPA (cyclophosphamide), and MTX (methotrexate). CAL 27 cells were resistant to treatment with VDS (vindesine sulfate), CDP (cis-platinum) or ACTD (actinomycin D). A culture submitted to the ATCC in December 1993 was found to be contaminated with mycoplasma. Progeny were cured by a 21-day treatment with BM Cycline.
ATCC complete growth medium: The base medium for this cell line is ATCC-formulated Dulbecco's Modified Eagle's Medium, Catalog No. 30-2002. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%. Temperature: 37.0°C
Subculturing:
Protocol: Remove spent medium, add fresh 0.25% trypsin, 0.53 mM EDTA solution, rinse and remove trypsin. Add fresh trypsin and let the culture sit at room temperature (or at 37C) until the cells detach. Add fresh medium, aspirate and dispense into new flasks. Subc*tion Ratio: A subc*tion ratio of 1:6 is recommended Medium Renewal: Every 2 to 3 days
