CD28抗體，Anti-CD28免疫組化實驗中常用的抗體為單克隆抗體和多克隆抗體。單克隆抗體是一個B淋巴細胞克隆分泌的抗體，應用細胞融合雜交瘤技術免疫動物制備。多克隆抗體是將純化后的抗原直接免疫動物后，從動物血中所獲得的免疫血清，是多個B淋巴細胞克隆所產生的抗體混合物。根據標記物的不同分為免疫熒光法，免疫酶標法，親和組織化學法，后者是以一種物質對某種組織成分具有高度親合力為基礎的檢測方法。這種方法敏感性更高，有利于微量抗原（抗體）在細胞或亞細胞水平的定位，其中*——抗*染色法zui常用。

實驗所用主要為組織標本和細胞標本兩大類，前者包括石蠟切片（病理大片和組織芯片）和冰凍切片，后者包括組織印片、細胞爬片和細胞涂片。 其中石蠟切片是制作組織標本zui常用、zui基本的方法，對于組織形態保存好，且能作連續切片，有利于各種染色對照觀察；還能*存檔，供回顧性研究；石蠟切片制作過程對組織內抗原暴露有一定的影響，但可進行抗原修復，是免疫組化中*的組織標本制作方法。
